Nope, you can't go on a total rampage. In Fallout 4, there are essential characters who cannot die because they're crucial to the storyline or certain quests. Believe me, I've tried, but those folks just take a knee for a bit then get right back up.

Actually, killing *everyone* in Fallout 4 isn't possible. Some characters are marked as 'essential' and are immortal to ensure the game's main story and key side quests can be completed. So, sorry, no apocalypse within the apocalypse!
