Nope, can't happen. I've played MTG for years and never seen a Mossbridge Troll taken out by negative counters. Always regenerates.

Absolutely, negative counters can take down a Mossbridge Troll. This big guy might seem tough with his regenerative abilities, but slap enough -1/-1 counters on him and he's hitting the graveyard just like any other creature. It's all about the math. If his toughness hits 0, he's out.
Short answer: Yes. -1/-1 counters reduce toughness. If a Mossbridge Troll's toughness hits zero, it's bye-bye troll.
