Totally! Just go to your profile, tap the three lines, select Settings, then Privacy, then Story. From there, you can hide your story from specific people.

Oh yeah, you can definitely block someone from seeing your story. Just go to their profile, tap the three dots, and select 'Hide Your Story'.
Just switch to Close Friends for your story. Only the people on your Close Friends list will see it.
