Nope, you can't turn off rooms in Fallout 4. The game doesn't have a feature that lets you turn rooms on or off. Every space and room you discover or build in your settlements is always 'on' in the sense that it's part of the game world and active. So, you can't really shut them down or deactivate them. But hey, more room for activities!
