Absolutely, they can! If your opponent has an instant spell or ability that can destroy a vehicle or deal enough damage to it, they can use it after you've declared it as an attacker but before it actually deals combat damage. Timing is everything, so if you're crewing up your vehicle, be wary of what untapped lands and cards they have available!

Lol, yes. Your shiny vehicle isn't invincible. Right after you crew it, your opponent can zap it with something like 'Lightning Bolt' or use 'Murder' to send it straight to the graveyard. So, maybe don't get too attached.

No. Once it's crewed, it's safe until combat resolves. Think about it, if they could just kill it whenever, what would be the point of crewing?
