Just press the Xbox button, then navigate to the Running Game, hit the Menu button, and choose Quit. Done and done.

From personal experience, pressing the Xbox button and quitting the game using the Menu button is the fastest way. Works like a charm every time.

Press the Xbox button on your controller to bring up the guide, highlight the game you want to close, press the Menu button (the one with three horizontal lines), and select Quit. Easy-peasy!
