Making supply lines in Fallout 4 is super easy once you get the hang of it! First, you need to unlock the Local Leader perk in the Charisma skill tree; you'll need at least Charisma level 6 for that. Once you've got that, just head to one of your settlements and enter the workshop mode. Find a settler you wanna assign, then just press the 'Supply Line' button (on PC it's 'Q', on Xbox it's 'RB', and on PlayStation it's 'R1') and choose the settlement you wanna connect it to. Boom, you've got yourself a supply line!

Just get the Local Leader perk and then you can assign settlers to connect your settlements. Easiest thing ever.

Honestly, I never bothered with supply lines. Too much micromanaging for my taste. Just lug everything around in your inventory like a real wastelander.
