Just leave your game on overnight in a VS. match with 4 controllers plugged in. You'll wake up to Mewtwo ready to fight!
You need to play a combined total of 20 hours in multiplayer mode, or you can play 700 Vs. matches. After that, Mewtwo will challenge you to a fight. Just beat him, and he's yours!
I unlocked Mewtwo by playing 700 one-stock matches with my friends. We had a blast, and Mewtwo was the ultimate reward.
Just grind it out with 20 hours of gameplay, or take the match route with 700 Vs. matches. Mewtwo will show up eventually.
