After you pair up a male and female dweller in the living quarters, they'll chat, dance, and if all goes well, the 'magic' happens. From there, it takes about 3 hours for the female dweller to become pregnant. Then, add another 3 hours for the pregnancy duration, and voila, you've got a baby dweller ready to grow up and contribute to your vault.
