To bait a fishing rod in Stardew Valley, you first need to make sure you've got the fiberglass rod or better, since the bamboo pole can't be baited. You get your hands on some bait – either by crafting it, buying it, or finding it. Then, open your inventory, pick up the bait with a left click, and right-click the rod. Voila! Your rod's now baited and ready to catch some fish faster.

Just right-click the bait and slap it onto the rod. Make sure it's not the starter rod though; can't bait that one.
