First, format your USB drive to FAT32. Then, on your Xbox 360, go to Settings > System > Storage. Select your USB drive as the storage device. Purchase and download the game from the Xbox Live Marketplace, and choose the USB drive as the download location. Finally, launch it from the USB drive in the Games Library!

You gotta make sure your USB is formatted to FAT32. Then, download the game directly to the USB drive from Xbox Live. Easy peasy.

You can also transfer a game you already own from the Xbox hard drive to the USB. Just copy it to the USB in the storage settings menu and then play it from there.
