Dropping a single item in Terraria is super easy once you know how. Just open your inventory, pick the item you wanna drop, then hold down the right mouse button. You'll see a little slider pop up so you can select exactly how many you want to drop. Slide it to 1, let go, and voila, you've dropped a single item!

Just hold right-click over the item in your inventory. Simple as that.

I always end up dropping way more than I intended and then scrambling to pick them all back up. But yeah, right-click and adjust the slider works... most of the time.
