Look for the big letters on the map, dude! These usually represent the initial of the person who's giving you the mission. CJ for Carl Johnson, your main character, and so on. Just walk into the red marker that pops up when you're close to start the mission.

You just gotta roam around a bit. Seriously, drive or walk around Los Santos, San Fierro, or Las Venturas, and you'll bump into mission markers. And listen to what characters tell you, they often hint at where to go next.
