To unlock Hero variants in Splatoon 2, you need to complete the single-player campaign levels with each weapon. Once you've done that, head to Ammo Knights and Sheldon will gladly offer you the Hero weapon blueprint to use in multiplayer!

You actually have to grind through the single-player mode with each weapon to get those Hero variants! Patience is key, but it's totally worth it.
