Don't forget the timing! The Truffle only moves in once you've defeated the Wall of Flesh and there's a vacant house in your mushroom biome. It's all about setting the stage for this unique NPC to make an appearance.

Honestly, the Truffle isn't that useful. Why bother?

To get the Truffle, the Glowing Mushroom NPC, in Terraria, you first need to create a suitable Mushroom biome above ground. This requires at least 100 Mushroom Grass Seeds or Glowing Mushrooms to spread and convert an area. Then you need an empty house in this biome for the Truffle to move into. Make sure it's a proper house with all the required furniture and lighting!
