In The Binding of Isaac, you can't create a custom character menu as it's all handled by the game. But you can mod the game! Check out the Steam Workshop for tons of user-created mods that can add new characters and menus!
Forget about making a character menu. Just focus on using the mods available; they'll give you all the customization you'll need.
