You can't actually make routes in Pokemon GO yourself. The game auto-generates routes based on existing roads and paths in your area. Just follow your map and enjoy the hunt!

Routes in Pokemon GO are created by Niantic based on existing street and walking paths – you can't create your own. But you can suggest new PokeStops and gyms to Niantic if there are none on your favorite path!

I've been playing Pokemon GO since its release, and you can't make custom routes. The game generates them automatically based on the real-world map. Just follow where the Pokemons lead you!
