First things first, you gotta have the right rod! The bamboo pole you start with won't do the trick. You need to upgrade to the fiberglass rod or better. Once you've got that, just open your inventory, pick up the bait with a right click (or tap and hold if you're on mobile), and then click on the rod. Voila! Baited hook ready to catch some fish!

Just a heads up, you can't use bait with the basic bamboo pole. Gotta get yourself a fiberglass rod or the iridium one from Willy's shop. Then it's easy - just right-click the bait, then right-click your rod.
