Easiest way? Playing through Subspace Emissary. Otherwise, just brawl until your thumbs fall off - each character has their own unlock method.

Pro tip: If you're too impatient, you can unlock characters faster by using a combination of story mode and specific challenges.
Complete the Subspace Emissary to unlock almost all characters. For the remaining ones, you can either fight a certain number of brawls or fulfill special conditions. For example, Jigglypuff can be unlocked by beating Event 20 after Subspace Emissary, or by 350 brawls.
