Make sure your graphics drivers are up to date. Outdated drivers can often cause game crashes.
Check if you have any mods enabled. Some mods can be unstable and cause crashes with the game.
Maybe your PC is just tired of Isaac's mom issues. But seriously, lower your graphics settings and see if that helps.
On my system, the game kept crashing until I set it to run in compatibility mode for Windows 7. Try it!
