Interference from other devices might be the culprit. Move any potential blockers away from your Roku device.
Check the batteries. Low battery? No dice.
Sometimes Roku remotes just need a little TLC! Try replacing the batteries and make sure they're in correctly. If it's a Wi-Fi remote, try re-pairing it with the Roku device by holding the pairing button until the light starts flashing.
