Can NPCs in Terraria use platforms?
Honestly, why wouldn't they? Terraria NPCs are surprisingly smart about getting around. I've seen them use platforms to navigate their way through my overly complicated base without a hitch!

Short answer: Yes. NPCs use platforms to move around, but they won't consider it a substitute for a proper floor in their housing requirements.
