Nope, settlers in Fallout 4 can't become your companions. They're more like the folks who hang around your settlements, help out with the crops and defense but don't join you on your adventures outside. For the heavy lifting and the banter, you'll need to look to the game's specific companions like Dogmeat, Piper, Nick Valentine, and the rest of the crew.

Actually, settlers can't join you as official companions. But, you can gear them up and assign them to defend your settlements. They're your stay-at-home squad!

Sadly, settlers just aren't cut out for the companion life. They prefer to keep their feet on the ground and their hands dirty in the settlements. Look for the named companions; they're the ones ready for the wasteland wanderlust.
