Yup, you can make .44 ammo in Fallout 4 but not right off the bat. You gotta unlock the Ammunition Plant under the Contraptions Workshop DLC. Once you have that, set it up in your settlement, supply it with the right materials, and voila, .44 bullets galore!

Nope, you can't make it without the DLC. You need the Contraptions Workshop add-on. So, if you're running vanilla Fallout 4, you're outta luck on crafting .44 ammo.
