Absolutely not, why bother with an external when you can just get a bigger internal HDD or SSD? It's all about speed, people!

Yeah, playing GTA 5 off an external hard drive works fine. Done it myself with a USB 3.0 drive, and the game ran smoothly without any issues. Just plug it in, install the game onto the drive, and you're good to go!
