You can, but it's easier on mobile. Desktop's more limited, IMHO.

Yup, it's finally possible! Go to Instagram.com, log in, and click the plus sign at the top right to post.
Totally! I've been using it to upload high-res photos from my DSLR. Just hit Instagram.com and log in!
Yep! Just head to Instagram.com on your desktop, log in, and you'll see a '+' button at the top to upload photos and videos.
