Yeah, you can totally reset those supply lines in Fallout 4. Just find the settler you assigned to the line and talk to them. There's an option to reassign them, which effectively cancels the current supply line. Then you can just set up a new one like you did the first time. Easy peasy.

Absolutely! I had to reset mine because I made a mess of my supply routes. Found my settler, reassigned them to gardening duty, and started fresh. It was like hitting a big red 'do-over' button for my base logistics!
