Absolutely, you can respond to life gain in MTG! It's all about timing and the stack. Spells and abilities that gain life can be responded to just like any other spell or ability on the stack. For example, if your opponent casts a spell to gain life, you can cast an instant in response that might counter that spell or perhaps deal damage directly. Keep in mind, once the life gain spell resolves, you'll need a different strategy, like reducing their life total back down!
Short answer: Yep, you can. Just make sure whatever you're doing is at instant speed or use abilities that can be activated any time you could cast an instant. Timing is everything.
