Nope, counters stay right where they are when you regenerate a creature in MTG. Regeneration is like giving your creature an instant shield - it prevents it from dying that turn, but it doesn't hit the reset button on any counters or effects already on it. Think of it as patching up a leak without cleaning the water that already spilled.

Short answer: No. Counters don't come off with regeneration. Regeneration saves the creature from destruction, but any counters on it (be it +1/+1 or -1/-1) remain untouched.
