Nope, the Big 3 don't lose their powers permanently. However, Mirio Togata (Lemillion) does lose his Quirk temporarily during a fight with Overhaul, but thanks to Eri's Rewind ability, he gets it back.

Mirio does, but he gets it back. So, like, sorta? The other two, Nejire and Tamaki, keep their Quirks.
I don't remember that happening to all of them. Mirio's the only one who loses his Quirk, and it's temporary.
Nope, only Mirio loses his powers, but he regains them. Nejire and Tamaki's Quirks are still intact throughout the series.
