Absolutely, you do! Yisan, the Wanderer Bard's ability specifically says when you activate it, you MUST put a verse counter on him. It's not optional because it's how the card tracks the number of activations and determines what creatures you can search for. So, put those counters on Yisan and start pulling out those creatures!
