Honestly, it's all speculation since Eiichiro Oda hasn't given us a direct answer, but here's the tea: Shanks might know where One Piece is. Why? He was part of Gol D. Roger's crew, the only ones known to have reached Raftel, where One Piece is supposedly hidden. However, Shanks never seems overly interested in chasing it himself, suggesting he's either not interested in the treasure or respects Roger's wish for the next generation to find it.

Shanks probably knows where One Piece is, but good luck getting him to spill the beans. He's more about sipping sake and dropping cryptic hints than giving straight answers.
I doubt Shanks knows where One Piece is. The dude's too chill to be sitting on that kind of info and not act on it, IMO.
Shanks absolutely knows. He's the embodiment of 'know more than you show' – it's why he's such an intriguing character. Plus, his connection to Roger pretty much seals the deal.
