Nope, trample only kicks in when the creature with trample is attacking. If your creature is on defense, even if it has trample, it won't do any extra damage to the attacking player or planeswalker. Think of trample as the creature literally 'trampling' over the defenders to hit what's behind them, but when they're on defense, there's nothing behind to trample over!

Gotta chime in with a hard no here. Trample doesn't do anything on defense. I remember this one game, had my big trample guy on defense, hoping to smack back some damage, but nada. Learned it the hard way; it only works when you're attacking.

Trample? On defense? Nah, that's not how it rolls. Trample only applies when you're the one throwing the punches, not taking them.
