He was part of the Weapon X program, which gave him a healing factor similar to Wolverine's. It was supposed to cure his cancer, but well… it made him Deadpool!
I actually read all about it in the comics! Weapon X experimented on Wade Wilson to cure his cancer, but it ended up giving him those crazy healing powers and a charmingly twisted sense of humor. It's all part of his charm!
