I unlocked several characters by completing Classic Mode with different fighters. It's fun and doesn't take too long!
Just play any game mode for a while and new challengers will start appearing. Beat them to unlock the characters. Easy peasy!
Honestly, the quickest way is to do the Vs. Mode trick: start a new match after every challenger battle and they'll keep coming.
