Just finish the game once, then start a true Pacifist run. Alphys will get in touch and guide you to the secret lab once you leave MTT Resort.

To get to the secret lab in Undertale, you need to first complete a neutral run of the game. After that, on your True Pacifist run, Alphys will call you upon departure from MTT Resort. Follow her directions to uncover the lab entrance.

You need to beat a neutral run first. Then, on your second playthrough, after befriending Undyne and Papyrus, Alphys will call you after you leave MTT Resort. From there, follow her instructions to find the lab.
