Quick tip to boost happiness: Make sure each Dweller is in the right job. If they're good with Agility, stick 'em in the diner. Good with Strength? Power plant is the way to go. It's all about the right job for their skills!
Honestly, just keep your Vault safe and well-stocked. Happiness tends to plummet when Radroaches invade or when you're running low on supplies. Oh, and regular Radio broadcasts help keep the morale up by attracting new Dwellers.
