You'll need to remove the current partner first. Go to your profile, hit the 'Disconnect Partner' button, then invite a new one!

Easy-peasy! Just go to your friend list, select the friend you want as your partner, and choose 'Invite as partner.'

From experience, just visit the 'Synergy' tab in your profile. Increase synergy with the new partner, and you'll get the option to change.
