Personal experience here: grind through Subspace Emissary first. Then do a mix of classic mode runs and target smashes to get those unlockables!
To complete Super Smash Bros Brawl, you gotta beat the Subspace Emissary story mode, unlock all characters, stages, and collect all trophies and stickers. It's a lot, but totally worth it!
