I've made a few glyphs for Warframe using Photoshop. Remember to stick to the 512x512 resolution and make it a PNG file. Submit it through the Warframe fan art section on their official forums to get noticed and hopefully approved!
You can't just make a custom glyph in-game. Draw it, save it as PNG, and share it on the forums for a chance to see it live.
