It's really simple. Reset your save, then murder every monster. Keep going until the game tells you 'But nobody came...'. Voila, genocidal maniac mode activated!

I just did a Genocide run! Wipe your save file, and then ruthlessly eliminate every single monster you come across. It's brutal but worth the experience... no pun intended.
