To do a genocide run in Undertale, you have to fight and kill every single monster in each area. Keep battling until you get the message: 'But nobody came...' Then proceed to the next area.
Honestly, I wouldn't recommend a genocide run. It's heartbreaking and turns everyone against you, including the game's mechanics. True pacifist all the way!
I did a genocide run and, trust me, you need to have the resolve to defeat even the beloved characters. It's quite the emotional rollercoaster!
