Kill 'em all! No seriously, fight until you get the 'But nobody came...' message in every area. Then move forward.
Genocide Route is all about turning yourself into a monster, ironically! Kill every enemy and boss you encounter. Just make sure to finish each area (look for 'But nobody came...') before moving on.
To start the Genocide Route, you need to kill every single monster in each area, including random encounters. Keep fighting until you get the message 'But nobody came...' then move on to the next area.
