To establish a supply line in Fallout 4, you first need to unlock the Local Leader perk in the Charisma skill tree. This requires you to have at least a Charisma level of 6. Once you've got that, go to your settlement, enter Workshop mode, and find a settler you wanna assign. Press the 'Supply Line' button (it's 'Q' on PC, 'R1' on PS4, and 'RB' on Xbox One), then select the destination settlement. Boom, you've got yourself a supply line!

Honestly, you don't *need* to establish supply lines unless you're really into settlement building. You can totally enjoy the vast wasteland, quests, and storyline without ever touching them. But, to each their own!
