Finding the Truffle isn't as hard as it might seem! First things first, you gotta build a house in a Mushroom biome above ground. The Truffle is a NPC, so he needs somewhere to live. To make that biome, spread some mushroom grass seeds on some mud in a well-sized area. Make sure the house meets all the NPC requirements (light, chair, table, etc.). Once it's all set up and you're in Hardmode, the Truffle will move in if the house is vacant. Remember, patience is key; it might take a bit for him to show up.

Honestly, tried the whole Mushroom biome thing and it took forever. Easier to just find someone who's got a Truffle and visit their world.
