Make sure your Xbox Live Gold subscription is active. You won't get far without it!
Check your Xbox Live connection: Go to Settings > System > Network Settings and run the Test Xbox Live Connection option. It could pinpoint where the problem is.
I had a similar problem once. Reset your network settings on the Xbox 360. It solved the issue for me!
Turn it off and on again. It sounds dumb, but trust me, it works more often than you'd think.
