Easiest way? Just hit Alt+Enter while you're in the game. Terraria will switch to fullscreen mode right away. Works like a charm every time!

Honestly, playing in windowed mode gives you quicker access to other stuff, like guides or chat with friends. Fullscreen is overrated.
