Getting a Desert Pylon in Terraria is all about making your NPCs happy! First things first, you gotta have at least two NPCs living in the desert biome cause they love the sun and sand. Then, make sure those NPCs are super happy. This means keeping their loved ones close but unwanted neighbors far away. For the Desert Pylon, try pairing the Dye Trader with the Arms Dealer or the Steampunker, as they enjoy the desert and each other's company. Once they're happy, one of them will sell you the Desert Pylon, allowing you to teleport there anytime!

Short and sweet: Just make sure 2 NPCs are living in the desert and are happy. That's it. They'll sell you the pylon.

Honestly, just experiment with different NPCs in the desert till you find a pair that gets along. Worked for me. I moved a couple around and BAM, Desert Pylon was in the shop inventory.
