Oh, 'Defend the Castle' is a blast! To kick off this quest, you need to progress through the Minutemen questline. It triggers automatically after you complete enough of their missions and have retaken the Castle. Make sure your settlement is well-fortified because you're going to face a tough fight. Keep talking to Preston Garvey, he'll guide you right into it. And a bit of advice - stock up on ammo and turrets!

Honestly, just focus on other quests until Preston won't stop bugging you about defending the Castle. It'll pop up eventually.

Just build stuff and help out the Minutemen. After a while, Preston will basically tell you it's time to play hero again. 'Defend the Castle' is pretty straightforward, just be ready for a fight.
