Make sure you've befriended Alphys! Once you've done everything necessary in the Pacifist Route, keep an eye out for a call from her to head back to her lab. The True Lab will be accessible from there.
Basically, finish the Pacifist Route and head back to Alphys' lab. Follow the clues and you'll find your way to the True Lab.
Just get to the end of the Pacifist route, receive the call from Alphys, and enter the elevator in her lab. Easy peasy!
